چکیده

Medicinal plants have been widely employed in treating various diseases for centuries and are still a major resource and repository that can be further exploited for a variety of infectious and non-infectious ailments. The popularity of medicinal herbs has grown significantly in recent years despite a dearth of information regarding their modes of action and continuing concerns over their efficacy. Hence there is a necessity to study various plants meticulously with respect to their biological properties. Recent efforts to elucidate the mechanisms of action of several anti-inflammatory herbs have focused on a class of compounds, sesquiterpene lactones, which are believed to be the active components of these herbal medicines. At present, the pharmacokinetics of sesquiterpene lactones is not well known. Parthenolide is a major sesquiterpene lactone isolated from medical herbs Tanacetum parthenium (commonly known as feverfew) and from Magnolia grandiflora. Feverfew has been traditionally used as a herbal medicine for the treatment of fever, arthritis, and migraine in Asia and Europe for centuries. Hence, the present study discusses various biological properties of parthenolide and its mode of action which can be used in drug designing processes to maximize efficacy and minimize side-effects.

Anti-proliferative effect and apoptotic induction of sesquiterpene lactone parthenolide in a human breast cancer cell line

Parthenolide is a secondary metabolite, which naturally occurs in the feverfew plant and is responsible for its healing power. The potential of parthenolide in inhibition of cancer cell growth, alone or in combination with other anti-cancer therapeutics, have been studied in several laboratories. In this study, the effect of extracted parthenolide on the expression of seven pro-apoptotic genes,...

Modulation of Cell Surface Protein Free Thiols: A Potential Novel Mechanism of Action of the Sesquiterpene Lactone Parthenolide

BACKGROUND There has been much interest in targeting intracellular redox pathways as a therapeutic approach for cancer. Given recent data to suggest that the redox status of extracellular protein thiol groups (i.e. exofacial thiols) effects cell behavior, we hypothesized that redox active anti-cancer agents would modulate exofacial protein thiols. METHODOLOGY/PRINCIPAL FINDINGS To test this h...

Oxidative stress-mediated apoptosis. The anticancer effect of the sesquiterpene lactone parthenolide.

The sesquiterpene lactone parthenolide, the principal active component in medicinal plants, has been used conventionally to treat migraines, inflammation, and tumors. However, the antitumor effects of parthenolide and the mechanism(s) involved are poorly understood. We found that parthenolide effectively inhibits hepatoma cell growth in a tumor cell-specific manner and triggers apoptosis of hep...

The sesquiterpene lactone parthenolide in combination with docetaxel reduces metastasis and improves survival in a xenograft model of breast cancer.

Parthenolide, a sesquiterpene lactone, shows antitumor activity in vitro, which correlates with its ability to inhibit the DNA binding of the antiapoptotic transcription factor nuclear factor kappaB (NF-kappaB) and activation of the c-Jun NH(2)-terminal kinase. In this study, we investigated the chemosensitizing activity of parthenolide in vitro as well as in MDA-MB-231 cell-derived xenograft m...

Mass detection for chromatographers: Benefits for the analysis of herbal medicines.

Feverfew (Tanacetum parthenium) is a medicinal herb that has long enjoyed traditional use as a remedy for migraine headache pain. The aerial parts of Feverfew (flowers, leaves and stems) contain parthenolide (a sesquiterpene lactone) which is believed to be responsible for the plant's biological activity. Dried feverfew aerial parts should contain approximately 0.2-0.5 wt.% parthenolide and adu...
